Glad to hear carb is working as expected.

Coil sounds like it’s worth a punt. What are the connections like as well.

Did you do new plugs with the new HT leads, either way are they correctly gapped?

The long screw just props open the throttle - you should see it rests on a little notch on a plate on the shaft. There is no voodoo or black magic like some screws on carbs, its mechanical. 

Sounds like your screw might have stripped - does it visibly change length as you screw it in/out?

Could be fuelling related. Two things to check to start with: 1) you don’t have any air leaks - spray carb cleaner or similar around all the various manifold joints, and also around the throttle spindles. For any future searchers, the answer was there was no need to bleed the clutch - just clamped off the big flexible braided feed hose, changed the cylinder, refilled with fluid and unclamped hose.

Not all covers are equal, they can (especially aftermarket ones) not seal that well.
